This is the first riding mission you can try right after purchasing Bullseye and it is pretty straightforward to complete in the 45 second time limit. There are two crates to collect and return to the green target circle, and both of them are in a straight line. The first is out to the east of the Shield Generator. Grab it and return in a straight line going west to drop it off.
Continue due west towards the Alien Village and grab the next crate. Spin around and head straight back to place it in the green target zone.

Horseback Supply Run
Mission Giver: Jessie Alien
Type: Fetch
Rewards: 250 Crystals / 100 Sparks
Like the previous Bullseye mission you must collect small supply crates and return them to the green target before time runs out. There is 1 minute and 30 seconds on the clock. That should be plenty of time if you remember to sprint with Bullseye instead of trotting along at a slower pace. The first crate is on the Shield Generator and a short ride on the lift plate will take you there in a jiffy.
The next one is on a ledge. The challenge here is that Bullseye can’t climb. To get to this crate you need to pass by it and go all the way to a ramp near the Alien Village. Follow that path to the edge of the cliff and make sure to grab the crate before leaping down.
The final crate is out in the Waterfall area and it will take a few tricky jumps to get to it. Be patient and slowly leap to each rocky formation to grab the crate. During this crate retrieval keep in mind that Bullseye can double jump like regular characters.
Jump back to the City and be sure to drop the last crate in the green circle. If you toss the crate or drop it out of the green ring it will not count.
Wii Horseback Supply Run
Wii Reward: 50 Crystals
Go to the teleporter at the Teleporter Dish and use it to warp to the first spot on the ledge near the entrance to Goo Valley. When you arrive, use the teleporter again to warp to the next location in the cliffside near the Shield Generator.
Drop down the ledge to the right and go down one more ledge to find the first crate near a small stream.
Perform the same double teleport warp to return to the same spot as before. Instead of dropping down the leftmost ridge, follow the edge of the rocky terrain farther out near the Character Vault and look down to the side to find the second crate.
Once again, repeat the double teleport trip, but this time go to the left. From here drop down two ledges to make your way next to the Challenge beacon to grab and retrieve the third crate.
